The initial patent for 44-ACO-DMT and a number of additional psilocin esters was issued on January 16, 1963 by Sandoz Ltd. through Albert Hofmann & Franz Troxler. In spite of this, 4-AcO-DMT is still classified as a psychedelic with a short history of usage. Before it was made available on the internet research chemical market as a gray area compound.

It is thought that 4-ACO-DMT’s partial agonistic activity at the 5-HT2A receptor accounts for some of its psychedelic effects. The function of these interactions and how they lead to the psychedelic experience, however, are still poorly understood.

What is 4-Aco-DMT?

It is believed that 4-AcO-DMT undergoes first pass metabolism in the body, where it is deacetylated into psilocin and then processed by the liver. This is based on reports that most users are unable to discriminate between these two substances when ingested, to the point where they are frequently thought to have similar subjective effects. Pharmacology

4-AcO-DMT is an ester of psilocin (4-HO-DMT), just like psilocybin (4-PO-DMT). The distinction between the two is that whereas 4-AcO-DMT is the O-acetylated form, hence the name O-Acetylpsilocin, psilocybin is the O-phosphorylated form, which is found naturally in mushrooms.Put otherwise, 4-AcO-DMT shares the same fundamental structure as psilocin and psilocybin, but it also has an acetoxy group attached to its indole ring.

Receptor binding

Receptor binding and metabolism of 4-AcO-DMT are little studied. Like psilocybin, it’s assumed to be a prodrug of psilocin—in this case, deacetylated to psilocin during first-pass metabolism by the stomach and liver. Considering this, it is reasonable to believe that 4-AcO-DMT acts in a manner similar to that of psilocybin and many other psychedelics by activating the 5-HT2A serotonin receptors.

Some people now think 4-AcO-DMT might be more than just a psilocin prodrug, though. Numerous anecdotal reports exist of individuals who inject or smoke 4-AcO-DMT and experience almost instantaneous psychoactivity (i.e., bypassing first-pass metabolism). This implies that it might have a unique pharmacokinetic profile, which could explain some of the variations in experiences between psilocybin and 4-AcO-DMT.However, it’s important to keep in mind that psilocybin that occurs naturally frequently coexists with other alkaloids that have distinct effects, like norbaeocystin and baeocystin, none of which are found in 4-AcO-DMT powders.
